The UIC Transgenic Production Service (TPS) provides a resource for members of the UIC community seeking to generate transgenic and gene-targeted (knockout) mice for their research activities. Services include (1) pronuclear microinjection of your DNA construct (transgene) into mouse zygotes for the production of transgenic founders, (2) injection of your targeted ES cell lines into blastocysts to produce chimeric mice, (3) re-derivation by embryo transfer to remove murine pathogens from your existing mouse strains, and (4) cryopreservation and related services. Investigators developing grant proposals that include the use of engineered animals are invited to contact the TPS Director for help with experimental design and for assistance in preparing proposed budgets for transgenic/knockout production and animal husbandry. We are happy to provide you with a letter stating our readiness to make transgenic and/or chimeric mice for submission to funding agencies. For knockout projects, consultation is also available to assist you in the generation of your targeted ES cell lines.

New Services!!! Gene Targeting in ES Cells

The TPS is pleased to announce the expansion of our services to include gene targeting via homologous recombination in ES cells.

We will electroporate your targeting vector into germline-competent ES cells, pick and freeze drug-selected clones and provide you with samples for genotyping (integration analysis). Identified clones will be thawed and expanded, and samples prepared for you to confirm correct targeting.

We will also test the targeted cells to determine that they are euploid and that they are mycoplasma-free. Suitable clones will be prepared for microinjection into blastocysts to produce chimeras.
